What is melanotan II?
Researchers at the University of Arizona created melanotan II in the late 1980s while looking for a way to tan skin without UV exposure, as a possible skin cancer prevention tool. It’s a cyclic peptide that’s more stable and more potent than natural alpha-MSH.
It binds several melanocortin receptors, including MC1R in skin and MC4R in the brain. Because it hits so many receptor types at once, its effects are broad, which is partly why later researchers built more targeted versions.
Melanotan II research
Small early trials found melanotan II darkened skin in volunteers. They also found unexpected sexual arousal effects, which led to the development of PT-141. Side effects reported included nausea, flushing and appetite loss.

Measuring and storage
It ships as a freeze-dried (lyophilized) powder in a sealed sterile glass vial. Before reconstitution, keep vials in the fridge at 2 to 8°C, or in the freezer for longer storage, away from light.
To reconstitute, let the vial reach room temperature, then add sterile diluent slowly down the side of the glass. Swirl gently and don’t shake it. As an example, adding 2 mL to a 10 mg vial gives 5 mg/mL, so every 0.1 mL holds 500 mcg. Write the date and volume on the vial.
Once mixed, keep it refrigerated and use it within the window your protocol allows. Most labs avoid repeated freezing and thawing of reconstituted peptide, because it can break the chains down.
