What is PT-141?

Bremelanotide is a cyclic seven-amino-acid peptide. It was developed from melanotan II after researchers noticed melanotan II had effects on sexual arousal.

Unlike melanotan II, it was refined to act mainly on MC4R and MC3R receptors in the brain, with less effect on skin pigment.

PT-141 clinical research

Palatin Technologies ran the RECONNECT phase III trials in premenopausal women with hypoactive sexual desire disorder. In 2019 the FDA approved bremelanotide as Vyleesi for that use. Nausea was the most common side effect, and some participants had temporary rises in blood pressure.

Measuring and storage

It ships as a freeze-dried (lyophilized) powder in a sealed sterile glass vial. Before reconstitution, keep vials in the fridge at 2 to 8°C, or in the freezer for longer storage, away from light.

To reconstitute, let the vial reach room temperature, then add sterile diluent slowly down the side of the glass. Swirl gently and don’t shake it. As an example, adding 2 mL to a 10 mg vial gives 5 mg/mL, so every 0.1 mL holds 500 mcg. Write the date and volume on the vial.

Once mixed, keep it refrigerated and use it within the window your protocol allows. Most labs avoid repeated freezing and thawing of reconstituted peptide, because it can break the chains down.
